Angiotensin-(1-7) stimulates the release of nitric oxide via phosphatidylinositol 3-kinase/Akt pathway in the nucleus tractus solitarii of rats
【摘要】：正Rationale:Angiotensin-1-7[Ang-(l-7)]via acting receptor Mas in the central nervous system is involved in cardiovascular regulation.Nitric oxide(NO) is implicated as an important modulator in the nucleus tractus solitarii(NTS),a key region involved in cardiovascular regulation. However,the signaling link involved in the interaction between Ang-(l-7) and NO release in the NTS remains unclear.Objective:The aim of the present study was to determine the role of phosphatidylinositol 3-kinase(PI3K)-Akt signaling pathways in mediating the effect of Ang-(l-7) on NO generation in the NTS of rats.Methods and Results:Ang-(l-7) injected into the NTS significantly increased NO generation and NO synthase(NOS) activity,which were abolished by the selective receptor Mas antagonist D-Alanine-[Ang-(l-7)](A-779) or the PI3K inhibitor LY294002.Western blotting analysis demonstrated that Ang-(l-7) stimulated the phosphorylations of Akt and NOS in the NTS,which was prevented in the presence of A-779 or LY294002.Microinjection of Ang-(l-7) into the NTS significantly augmented the response of blood pressure,heart rate,and renal sympathetic nerve activity to blockade of NOS,which was abolished by pretreatment with A-779 or LY294002.Pretreatment with Ang-(l-7) significantly enhanced the cardiovascular inhibition evoked by the NO precursor L-arginine(2 nmol) injected into the NTS.In addition,chronic intracerebroventricular infusion of Ang-(l-7)(200 pmol/day, 3 days) also increased NO release in the NTS in a Mas-dependent manner.Conclusion:We suggest that Ang-(l-7) stimulates the release of nitric oxide in the NTS,which is dependent on the Mas-PI3K-Akt-NOS signaling pathway.
